Prov Medium Derivatives

These media contain less Alkaline Soil Extract and are supplemented with NH₄Cl. (To make 50 mM NH₄Cl Stock Solution dissolve 2.68 g NH₄Cl in 1.0 L dH₂O).

Prov50:
Prepare as for Prov Medium, but use only 10.0 ml Alkaline Soil Extract. Add 1.0 ml 50 mM NH₄Cl Stock Solution. Bring final volume up to 1.0 L with filtered seawater and autoclave after all additions.

Prov50+Org:
Dispense Prov50 into desired culture vessel, add Organics Stock Solution then autoclave. Organics Stock Solution should comprise 5% of the final volume of this medium.

Organics Stock Solution:
Sodium Acetate 1.0 g
Glucose 6.0 g
Sodium Succinate 6H₂O 3.0 g
Neopeptone 4.0 g
Bacto Tryptone 4.0 g
Yeast extract 2.0 g
Distilled water to 1.0 L

Dispense in small aliquots and autoclave.

Prov50+Rice:
Add one grain of white rice to a tube of Prov50 then autoclave. The addition of rice encourages bacterial growth, and is helpful when maintaining mixotrophic and heterotrophic strains.

Prov100:
Prepare as for Prov Medium, but use only 10.0 ml Alkaline Soil Extract. Add 2.0 ml 50 mM NH₄Cl Stock Solution. Make final volume up to 1.0 L with filtered seawater and autoclave after all additions.

ProvM (75%SW):
This medium is used to maintain organisms from brackish environments (approximately 24 ppt salinity). Mix 750 ml filtered seawater and 200.0 ml dH₂O. Prepare as for Prov Medium, but add only 5.0 ml Alkaline Soil Extract. Make final volume up to 1.0 L with dH₂O, then autoclave.
